    Clinical Background

Approximately two-thirds of breast cancers express estrogen receptor alpha. These estrogen-receptor-positive tumors depend on estrogen-receptor signaling for growth and progression. Selective estrogen receptor modulators (SERMs) and selective estrogen receptor down-regulators (SERDs) are endocrine therapies that block estrogen-receptor function or signaling. Some SERMs also have estrogenic effects on bone and may help prevent bone loss, improve bone mineral density, and reduce vertebral-fracture risk.
