Teprotumumab binds to and blocks activation of the insulin-like growth factor-1 receptor (IGF-1R), and is used to treat inflammatory orbital manifestations of TED such as proptosis, diplopia, eyelid retraction, and soft tissue swelling. Randomized trials demonstrated rapid and clinically meaningful reductions in proptosis and improvements in clinical activity and diplopia versus placebo, with phase 3 studies reporting substantially higher proptosis responder rates and a Phase 4 trial showing statistically significant proptosis reduction at Week 24.
